If you want duo, then go with the E8400. Otherwise, get the Q6600. Depends what you gonna do with it.
I'd say dual, unless you're gonna do much multithreading. Most games run better on dual.

The idea of any hi-fi system is to reproduce the source material as faithfully as possible, and to deliberately add distortion to everything you hear (due to amplifier deficiencies) because it sounds 'nice' is simply not high fidelity. If that is what you want to hear then there is no problem with that, but by adding so much additional material (by way of harmonics and intermodulation) you have a tailored sound system, not a hi-fi. - Rod Elliot, ESP

The quad is more 'future proof'.
The dual has more raw grunt.

As others have said, what are you upgrading from and what do you want to do with it?
If you're running multiple cpu-intensive apps, then the quad will be better - and will be better for future multi-threaded games.
Also if the low grunt of the quad isn't going to be a huge leap up from what you've got, I'd hold off for now - wait for quad prices drop a bit - so you can get one with more grunt - particular if you're say upgrading from a 2.xxGhz dual.
That goes doubly so if the dual ain't a big step up on what you've currently got.
