There are plenty of non-fliers who became astronauts, but without knowing what the normal crew composition will be when you get to that point (20-odd years from now), the safest way to ensure you are competitive is to become a military pilot (USAF, USN, or USMC...USA doesn't have high-performance aircraft). Then become a test pilot in any of the three branches.
Take a look at this guy's bio to get an idea of what's required.
